Use of in vivo and in vitro tests to evaluate toxicity of chemical substances and drugs

This course describes the pharmacological basis of therapeutics. Each chapter begins with a short description of pathophysiology. Therapeutic targets and the drugs acting on them are highlighted. Novel directions in therapeutic strategy may also be described.

To evaluate toxicity of chemical substances and drugs, and before human use, several experimental steps must be done :
- micro-organism tests : speed and cheap, but with a lot of disadvantages (cell wall, none micro-oranite, specificity of ADN repair mechansisms, etc...);
- eukaryotic cells tests : allow to control exactly the concentration used but not the study of influence of metabolic activation/inactivation (important in the study of cancer);
- in vivo tests : results more sensitive and easier to transpose to human; the choice of a species is the "key" choice ; it is determinated by taking into account metabolic process and detoxification mechanisms.

At the end of this course, the students are supposed:
- To know the main targets of drugs that are used in therapeutics, - To understand why it is interesting to act on such targets, - To be able to give a few examples of drugs that are used clinically, - To describe innovative strategies for the discovery of new drugs.

Having had success in "Toxicological precepts" of the former academic year
Notions of general pharmacology, physiology and pathophysiology.
