Learning unit contents

The theoretical course is divided into 10 chapters. The first two chapters are devoted to the organization of genes and genomes and to their replication and expression (transcription, translation). The three next chapters concern gene transmission, recombination and interaction between genes. Chapters 6 and 7 describe the types of mutations. Chapter 8 describes genetics of phages and bacteria. Chapter 9 is devoted to organelle genetics. Chapter 10 briefly describes the technology of recombinant DNA.

Practical courses: one afternoon is devoted to bacterial conjugation in Escherichia coli (interrupted conjugation) and the other one to intragenic complementation of arg- markers in the unicellular green alga Chlamydomonas reinhardtii.


Bacterial conjugation: conjugation is made between a donor strain harboring a fertility plasmid and a recipient strain. Conjugation is interrupted at defined times. Transfer of plasmid is detected by plating bacteria on growth media supplemented with antibiotics.


Intragenic complementation: activity of the argininosuccinate lyase is measured before and after a heat shock in a wild-type haploid strain, in mutant haploid arg- strain and in a diploid strain showing intragenic complementation at the arg7 locus.


At the end of the theoretical course, one afternoon is devoted to the resolution of problems of genetics.

For 1er session, written examination for problems of genetics and oral for theory. For 2nd session, written examination only for both problems of genetics and theory.
 
En cas d'examen à distance, l'examen sera oral à distance et comprendra une partie consacrée à la résolution de problèmes et une partie consacrée à la théorie.
